Is it possible to overdrive a preamp tube too much (ie: too much overdrive/distortion) to cause it to the tube to fail? A new preamp tube I just put in V1 is starting to sound fuzzy/more distorted than it did when I first put it in (a few weeks ago), especially when I run an OD pedal into it. Any thoughts? I do do run a few high gain pedals into the front end....

Nope. Tubes fail for a lot of reasons, but unless the amp was designed poorly, then the tube shouldn't fail for that reason.
