For Telecaster neck pickups that are not mounted to the pickguard, do the mounting screws go directly into the wood or are there supposed to be some kind of threaded sleeves mounted into the wood that the screws can screw into?

Also are springs or plastic tubing used when mounting a Tele neck pickup in this fashion?

the screws should have either a rubber sleeve or springs, but the screw goes right into the wood holes.

If you have never done this before, be careful. It is a b1tch to line everything up. Go slow and triple check. Use a drill press to drill te holes to ensure proper adjustment ability.

Yeah and make sure you don't let the drill go out the back of the body. I've been there done that...

I've never done that but do have a very nice little hole in the headstock of one of my Tele's from installing tuners.

Bill Callaham and Lindy Fralin have a small wood screw, #2 I think, that is a quarter inch shorter than the factory length neck pup mount screw. (3/4 ths inches long). More than adequate to do the job, and the risk of "misadventure" is next to nothing.

I drill just a guide hole myself, the wood is more than soft enough so there is no chance of drilling through the back and I use a very small bit going in about 1/4 inch. works great for me.

I realize you'd lose adjustability, but would someone hear a tonal difference when mounting the neck pickup directly into the wood, as opposed to mounting with screws into the pickguard? (pickup heights being equal, of course.)

Build them both ways and my ears cannot hear a difference. Now let the debate begin!:munch
