Would a Two Rock Classic Type III benefit from a Dumbleator?

Even if I don't use the loop? I run a W/D/W rig so I don't use an effects loop.

What kind of changes could I expect to hear with a Dumbleator hooked up?

Even if you don't loop back to the amp, the effect(s) you take out of the amp to run the WDW rig can affect the tone of both the amp and the effects. The buffering effect that an outboard loop adds, would cure that. Your dry tone would remain as it is, and effect tone would likely be better.

Do you mean if I just use the 'send' only to feed my effects? I actually use the line out from a Hotplate, so the loop isn't used at all.... I should have clarified.

I did try just running a 1' patch cord in the loop but didn't hear any difference.

I've got TR Custom Proto #2 (fine indeed). I has a built in dumbleator. I've been running with it on mostly because it has been so handy to have a MV on the front panel. I always has an issue with the way the Dirty rhythm responded. It never felt quite right. Not organic or natural is the only way I can describe it. Always loved everything about the cleans and dirty single notes lines.

Due to my twiddling 2 year old, I had to fix every setting and could not get it to sound right. I flipped off the Loop and the amps response changed. Everything seems to jump out of the amp just a little faster. The dirty rhythm came alive and I am much happier with the way the amp feels.

I will say that the amp without the loop on, is a little brighter overall but I believe at least part of that is due to my fiddling (The controls are pretty darn close to what they've always been so Part of it is the loop as well). I can't say one is better than the other but I prefer the loop off.

i think it has been said, when you get to the level of quality of these amps, the differences are often subtle, But that is eaxctly what you are paying for. I hope for your sake, you get a change to try before you buy in this case.
